چکیده

background: during the initial phase of an infection, there is an upregulation of inducible nitric oxide synthase in the macrophages for the production of nitric oxide. this is followed by the recruitment of polymorphonuclear leukocytes (neutrophils) which release arginase. arginase competes with inducible nitric oxide synthase for a common substrate l-arginine. objective: to investigate whether the entry of neutrophils and release of arginase can interfere with nitric oxide production from stimulated mouse macrophages. methods: neutrophils were isolated from human blood and stimulated with cytodex-3 beads. cultured macrophages were stimulated with lipopolysaccharide and interferon gamma with or without n (g)-nitro-l-arginine methyl ester or n (omega)-hydroxy-nor-l-arginine. measurement of no2 - /no3 - and urea were done using the spectrophotometer. results: a significantly higher level of nitric oxide production from stimulated macrophages was observed compared to control. there was a decrease in nitric oxide production when stimulated macrophages were treated with the supernatant from activated neutrophils (p

Inhibitory effect of sulphated polysaccharide porphyran on nitric oxide production in lipopolysaccharide-stimulated RAW264.7 macrophages.

Porphyran, extracted from an edible red alga (Porphyra yezoensis), is a sulphated polysaccharide with a wide variety of biological activities including anti-tumour, antioxidant and immuno-modulating activities. In this study, we examined the effect of porphyran on nitric oxide (NO) production in mouse macrophage cell line RAW264.7 cells. Although no significant activity of porphyran to induce N...

Effect of quercetin on the production of nitric oxide in murine macrophages stimulated with lipopolysaccharide from Prevotella intermedia

PURPOSE Nitric oxide (NO) is a short-lived bioactive molecule that is known to play an important role in the pathogenesis of periodontal disease. In the current study, we investigated the effect of the flavonoid quercetin on the production of NO in murine macrophages activated with lipopolysaccharide (LPS) from Prevotella intermedia, a pathogen related to inflammatory periodontal disease, and t...

Synergistic Effect of LPS, IFN- and Iron on Apoptosis of Balb/c Mice Macrophages Following Nitric Oxide Production

Objective(s) Previous studies have demonstrated that the nitric oxide (NO) dependent death of murine peritoneal macrophages activated in vitro with IFN-g and LPS is mediated through apoptosis. In the present study, we investigated the synergistic effect of LPS, IFN-g and iron on NO production and apoptosis. Materials and Methods After determination of iron cytotoxicity, the peritoneal macrop...

Comparative nitric oxide production by LPS-stimulated monocyte-derived macrophages from Ovis canadensis and Ovis aries.

Bighorn sheep are more susceptible to respiratory infection by Mannheimia haemolytica than are domestic sheep. In response to bacterial challenge, macrophages produce a number of molecules that play key roles in the inflammatory response, including highly reactive nitrogen intermediates such as nitric oxide (NO). Supernatants from monocyte-derived macrophages cultured with M. haemolytica LPS we...
